~(99)Tc~m标记N_3O_3-Schiff碱类配合物的定量构效关系研究

摘要: 用分子力学、半经验量子化学的ZINDO/1方法对系列99Tcm标记N3O3 Schiff碱类配合物的结构进行了优化,作了电子结构计算。并将这些参数对心肌初始摄取值进行多元线性回归分析,得到了回归方程。利用所得方程对该类配合物的心肌摄取机理进行了讨论,提出了该类配合物的心肌摄取机理可能为被动扩散机理。具有低极性的配合物比较容易通过被动扩散机理进入心肌细胞中,在最大正电荷的影响下"陷落"在线粒体中。Abstract: The geometries of \~+ complexes are optimized by the semi-empirical quantum mechanics method——ZINDO/1 and many physical and chemical descriptors are obtained. Step multiple regression between these descriptors and heart initial uptake is done and an equation is obtained. New uptake mechanism is proposed: the complexes with low polarity might be passively transported into the myocardium cell, and would be "trapped" by proteins with negative charge on the membrane mitochondria because of the Q~+max) of the complexes.
