高效液相色谱法分析高比活度γ-~(32)PATP和α-~(32)PdATP
ANALYSES OF γ-~(32)PATP AND α-~(32)P dATP AT HIGH SPECIFIC ACTIVITY BY HIGH PERFORMANCE LIQUID CHROMATOGRAPHY

摘要: 本工作叙述了高比活度的γ·~(32)PATP和α·~(32)PdATP的放化纯度分析,研究了dATP和ATP的分离因子与流动相中磷酸盐浓度的关系。在YWG-R_4NCl阴离子交换柱上,用磷酸盐溶液梯度淋洗或乙醇-磷酸盐缓冲液作流动相分离了A,dA,AMP,dAMP,ADP,dADP,ATP和dATP。ATP和dATP在紫外检测器上用峰高法进行了定量测定,ATP的最小检测量为1ng。用834色谱微处理机进行放化纯度定量测定,标准偏差为0.65%。本方法用于高比活度γ·~(32)PATP和α·~(32)PdATP标记过程中的样品分析和纯化后的产品分析。Abstract: In this paper analyses of radiochemical purity forγ-32PATP and α-32PdATP at high specific activity are described. The relationship between separation factor a and concentration of phosphate buffers in eluant is investigated. The separation is carried out on an YWG-R4NC1 amino column, using a gradient or isocratic elution. Detection is selected at 254 nm. Radioactivity is measured by a plastic scintillation flow detector.
