希土放射性核素通过TTHA标记I_gG和单克隆抗体——Ⅰ.标记方法研究

    LABELLING OF TTHA-COUPLED IgG AND MCAb WITH RARE EARTH RADIONUCLIDES——I. EXPERIMENTAL METHOD

    • 摘要: 本文提出了用三乙撑四胺六乙酸(TTHA)环状酸酐(CTTHAA)作为双功能螯合剂,预先连接到免疫球蛋白G(IgG)上,再络合希土放射性核素,标记IgG和单克隆抗体的方法。并对影响标记的因素,如金属与CTTHAA摩尔比,pH和反应时间作了详细研究。标记物RE-TTHA-IgG仍保持免疫反应活性。

       

      Abstract: A method for labelling G-immunoglobulin and monoclonal antibody with rare earth radionuclides is reported. In this labelling process cycloanhydride of TTHA is employed as a bifunctional chelating conjugate to make prior linkage of TTHA to globulin, then followed by the metal chelation.The influencing parameters of labelling globulin with rare earth, such as metal to TTHA ratio, pH value and labelling time are investigated in detail. Through the whole labelling process the immunoreactivity of the labelled compound is well preserved.
